HaloTag is a self-labeling protein tag engineered via site-directed mutagenesis from haloalkane dehalogenase (DhaA) of Rhodococcus rhodochrous. It consists of 297 amino acids with a molecular weight of approximately 33 kDa. Unlike short peptide tags such as His, HA and Myc, HaloTag can form stable and irreversible covalent bonds with chloroalkane ligands. When conjugated with fluorophores, biotin or resin-modified ligands, it enables a full spectrum of applications including live-cell imaging, protein purification, protein-protein interaction capture, pulse-chase assays and kinetic analysis of protein degradation. |
